Bob Seger said in an interview the song was inspired the doings at a club on Ann street in Ann Arbor Michigan where he went to High School. Ann street was one block off of Main Street.

The Main Street that the song is talking about is Main Street Columbus, Ohio back in the early seventies and eighties used to be the largest street for clubs on route 40 and the US there were so many strip clubs It was amazing believe it or not the playboy club on by Hugh Hefner who is also in this town.

"Main Street" is a song about a middle aged man reminiscing about a cute young stripper he had a major crush on, back when he was a shy teenager.
Most of the other strippers had a hard edge- they'd learned to be tough and to keep customers at arm's length. But this one stripper seemed much younger, sweeter and more innocent than the others. The narrator of the song imagined that, somehow, he might actually be able to talk to her and form a real relationship with her. But he never had the guts to actually approach her.
Many, many years later, the narrator admits he still thinks about that girl, and sometimes wanders down Main Street, where she used to dance, and tries to picture her.
